Introduction
Usages: For isolation and enumeration of coagulase-positive staphylococci in foodstuffs.
Principle: Tryptone, beef extract powder and yeast extract powder provides carbon and nitrogen sources, vitamins and growth factors; sodium pyruvate and glycine to stimulate the growth of Staphylococcus aureus; lithium chloride and potassium tellurite inhibit the non-staphylococcal microorganisms; containing lecithinase staphylococcal colonies produce degradation yolk make transparent circle, while the role of the lipase produced an opaque precipitate ring; coagulase-positive staphylococci can restore potassium tellurite and produce black colonies; agar is medium coagulant.
Formulation(per liter): Pancreatic Digest of Casein 10g Beef Extract 5g Yeast Extract 1g Sodium Pyruvate 10g Glycine 12g Lithium Chloride 5g Agar 20g Final pH7.0 ± 0.2
How to use: 1.Suspend 63g in 950ML of distilled water , stirring heated to boiling ,autoclave at 121℃ for 15 minutes. 2.Diluted and treated samples.

For selective isolation and culture of Staphylococcus aureus.
Principle:
Peptone and beef extract powder provides carbon, nitrogen, vitamins and minerals; D- mannitol to fermentable sugars; higher levels of sodium chloride to provide a higher osmotic pressure, suppress most non-staphylococcal microorganisms ; phenolsulfonphthalein as pH indicator; agar is medium coagulant. Typical pathogenic staphylococci (coagulase positive) D- mannitol produce acid fermentation and produce yellow colonies with a yellow halo, typically non-pathogenic Staphylococcus unfermented D- mannitol to form red colonies.

Usages: For isolating lactose-fermenting Gram-negative enteric bacilli.
Principle: Peptones are sources of nitrogen and other nutrients. Lactose is a fermentable carbohydrate. When lactose is fermented, alocal pH drop around the colony causes a color change in the pH indicator (neutral red) and bile precipitation. Bile salts,bile salts no. 3, oxgall and crystal violet are selective agents that inhibit growth of gram-positive organisms. Agar is the solidifying agent.

How to use: 1.Suspend 50 g in 1 L of distilled or deionized water. Heat to boiling to dissolve completely. Autoclave at 121°C for 15 minutes.
2.Transfer 1 mL of Soybean–Casein Digest Broth to 100 mL of MacConkey Broth, and incubate at 42 to 44 for 24 to 48 hours. Subculture on a plate of MacConkey Agar at 30 to 35 deg.C for 18 to 72 hours.
