OverView
ArcticZymes’ Shrimp Alkaline Phosphatase (SAP), including its recombinant version (rSAP), is the gold standard and the first heat-labile, all-purpose alkaline phosphatase on the market.
Originally isolated from Pandalus borealis (Arctic shrimp), it has been purified from a recombinant source since 2010. Unlike other alkaline phosphatases, rSAP can be fully inactivated by a short heat treatment of 15 minutes at 65°C.
This added convenience through complete heat inactivation has made SAP one of the most sold DNA modifying enzymes.
For added flexibility, especially when lyophilisation may be desired, rSAP is also available in a Glycerol FREE format. First launched in 1993, SAP’s unique features continue to make it a valuable tool in various applications.

Norovirus is known to cause acute gastroenteritis. It is a small (27-38 nm), round, nonenveloped RNA virus belonging to the Caliciviridae family and is responsible for over 80% of non-bacterial outbreaks of gastroenteritis in the world. It affects individuals of all ages, with a distinct seasonal link to winter. It has a genome of 7.6 kb that is positive sense and has a single stranded linear confirmation. It encodes a major structural protein (VP1) of about 58 to 60 kDa and a minor capsid protein (VP2). Transmission occurs predominantly through ingestion of contaminated water, food and airborne transmission, as well as contact with contaminated surfaces. The ease with which norovirus is transmitted and the low infectious dose required to establish an infection results in extensive outbreaks in numerous environments, such as hospitals, hotels and schools. There is no antiviral drug available to treat this infection and little is known about its pathogenicity. However, it has been observed that the virus can be taken up by enterocytes where translation of viral nonstructural proteins can occur; it damages and alters intestinal microvilli, leaving them blunt and broadened, thus inhibiting absorption; it causes crypt cell hyperplasia and also leads to apoptosis of enterocyctes. An incubation period of 24-48 hours is usual. Infection is characterized by the acute onset of nausea, vomiting, abdominal cramps, aching limbs, raised temperature and diarrhoea that generally last for about 48 hours. However, more severe and prolonged infection may be observed in children and the elderly. There are five recognized norovirus genogroups, of which three (GI, GII, and GIV) are known to affect humans and, since 2002, variants of the GII.4 genotype have been the most common cause of norovirus outbreaks. There have been 31 different genotypes identified within the genogroups, with a wide degree of genetic variability present even within each genotype.

About Inclusion Bodies
Bacteria are widely used for the expression of different proteins. However, 70-80% of the proteins expressed in bacteria by recombinant techniques are typically contained in insoluble inclusion bodies (i.e., protein aggregates). The protein of interest found in these subcellular structures is often inactive, due to incorrect folding. The production rate of recombinant proteins stored in inclusion bodies is invariably higher than those synthesized as soluble proteins. The reason behind this is thought to be the resistance of insoluble proteins to proteolysis by cellular enzymes. In addition, separation of insoluble recombinant proteins in inclusion bodies is considerably easier than that of soluble proteins. These factors have been the major influences favoring scale-up of high-value proteins using bacterial fermentation for example. Procedures for the purification of the expressed proteins from inclusion bodies are often labour-intensive, time-consuming and not cost-effective. This kit provides the essential reagents for cell disruption, inclusion body solubilization and purification using spin column chromatography – all optimized to work together thereby simplifying the process and saving a tremendous amount of time and cost.
The Cell Lysis Reagent and IB Solubilization Reagent should be stored at 4°C upon receipt of this kit. All other solutions should be kept tightly sealed and stored at room temperature. This kit is stable for 2 years after the date of shipment. Once opened, the solutions should be stored at 4°C when not in use except for the pH Binding Buffers (Acidic and Basic). Some precipitation will occur with 4°C storage. This precipitation should be dissolved with slight heating to room temperature before using.
