Adenoviral vectors are useful tools for both in vitro and in vivo gene transfer, and oncolytic viruses based on adenovirus are highly promising for cancer treatment. Norgen’s Adenovirus Purification Kit provides a fast and simple procedure for concentrating and purifying adenoviral vectors from cell lysate and cell culture media. Purification is based on spin column chromatography using Norgen’s proprietary resin as an ion exchanger. Contaminating cellular debris is largely removed from the sample via a centrifugation step, while contaminating DNA and RNA is reduced using enzymatic digestion. Adenoviral vectors purified in this manner are highly active for use in transduction experiments.
Norgen’s Adenovirus Purification Kit contains sufficient materials for 15 preparations (33.5 mL per prep of supernatant or a total of 500 mL of supernatant input). Approximately 1 mL of cell pellet can be purified per prep, up to a maximum of 15 mL of cell pellet in total for the entire kit.

HL-SAN Nuclease should be stored at -20°C upon arrival. Elution Buffer P should be stored tightly capped at 4°C upon arrival. All other solutions should be kept tightly sealed and stored at room temperature. This kit is stable for 2 years after the date of shipment.

Safety Features
1. User-Friendly Design: The chamber lid and tabletop are covered with heat-insulating resin materials, providing both protection against burns and an aesthetically pleasing appearance. The upward-opening flip-top door saves space, while the rounded corners prevent injury. The specially designed steam discharge and collection system minimizes steam burn risks and enhances operational convenience.
2. Interlock Mechanism and Safety Protection: The safety interlock system is designed to be more sensitive and secure. The chamber lid cannot be opened if there is pressure or steam inside the chamber. The lid can only be opened when the pressure gauge reads “0” MPa. The sterilizer will not start heating if the chamber lid is not properly locked. These features enhance the overall safety of the machine.
3. Over-temperature and Heating Protection: The system continuously monitors the temperature inside the sterilization chamber. If the temperature becomes too high or heating is abnormal, power will be automatically cut off to prevent overheating.
4. Automatic Fault Detection System: The machine performs an automatic self-check when powered on. If a fault occurs during operation, an audible alarm will sound, and a fault code will be displayed.
5. Lid Closure Check System: The system automatically detects whether the chamber lid is securely locked. If the lid is not properly locked, the sterilizer will not start.
6. Leakage, Over-current, and Short-Circuit Protection: If there is leakage in the heating elements or other components, or if over-current or short-circuit conditions occur, the system will automatically cut off power to prevent user injury.
7. Water Shortage and Independent Dry-Burning Protection: Equipped with a low water level sensor and independent dry-burning protection device, the system can monitor the water level in the chamber in real-time to prevent dry burning and avoid false readings. If the low water level sensor fails, the overheat and dry-burning protection system will automatically activate, cut off power to the heating ring, and sound an alarm to ensure safety.
8. Dual Pressure Protection: The system includes both a mechanical safety valve and an electronic over-pressure protection system. If pressure becomes abnormal, the system will relieve pressure and cut off power while sounding an alarm.
9. Back end Safety Testing: The system can be periodically tested for safety and protective measures.
10. Over-pressure Testing: Regular testing ensures that the instrument’s over-pressure protection system can automatically cut off power or that the safety valve can open to relieve pressure.
11. Performance Testing: The instrument’s safety performance has been tested by national authorized inspection agencies, and a test report is available.

Mastitis is the single most costly disease of dairy cattle resulting in the reduction of milk yield and quality. The inflammation of the utter is mainly caused by infection of various bacteria. Streptococcus uberis is a gram-positive bacterium that is known worldwide as an environmental pathogen responsible for a high proportion of cases of mastitis in lactating cows and is also the predominant organism isolated from mammary glands during the non-lactating period. Often it is resistant to treatment and causes persistent high somatic cell counts without clinical mastitis.

All kit components should be stored at -20°C upon arrival. Repeated thawing and freezing (> 2 x) of the Master Mix and Positive Control should be avoided, as this may affect the performance of the assay. If the reagents are to be used only intermittently, they should be frozen in aliquots.

Principle and Interpretation
Tryptone, proteose peptone, meat extract, yeast paste powder provide nitrogen sources, vitamins, amino acids, and growth factors; sodium chloride can maintain a balanced osmotic pressure; sodium dihydrogen phosphate and potassium dihydrogen phosphate as buffering agents; esculin are fermentable sugars; lithium chloride and other antibiotics can inhibit Gram negative bacteria and most Gram positive bacteria.

Preparation
Suspend 57.4g in 1 L of distilled or deionized water. Heat with frequent agitation and boil to completely dissolve the powder. Distribute into flasks. Autoclave at 121℃ for 15 minutes. Cool to 45-50℃,aseptically add the contents of 1 vial of Fraser Selective Supplement (SR0120) to each 225 mL of base to prepare FB1 solution; Add one reagent (SR0130) to each100 mL of FB2 culture medium to prepare FB2 enrichment solution.

Cultural characteristics observed after incubation at 35-37°C for 46~50 hours
