PAX-8 is a member of the paired box (PAX) family of transcription factors, which are key regulators in early development. This protein plays a role in development of thyroid follicular cells and the expression of thyroid-specific genes, with mutations in the PAX-8 gene linked to thyroid follicular carcinomas, atypical thyroid adenomas, and thyroid dysgenesis. The PAX-8 protein is expressed in simple ovarian inclusion cysts and non-ciliated mucosal cells of the fallopian tubes, but is absent from normal ovarian surface epithelial cells. PAX-8 is also not expressed in normal lung or lung carcinomas. Reports have associated PAX-8 expression with renal carcinoma, nephroblastoma, and seminoma, and have indicated PAX-8 as a useful marker for renal epithelial tumors, ovarian cancer, and for differential diagnoses in lung and neck tumors. Anti-PAX-8 can be useful in determining the primary site of invasive micropapillary carcinomas of ovary from bladder, lung, and breast, when used in adjunct with a panel of organ-specific markers such as uroplakin, mammaglobin, and TTF-1.

The ExcelTaq™ Blood Direct DNA Polymerase is designed for amplifying targeted DNA directly from whole blood, eliminating the need for a lengthy DNA isolation process. ExcelTaq™ Blood Direct DNA Polymerase is highly tolerant in the presence of PCR interfering/ inhibiting substances in blood, such as IgG, hemoglobin, and lactoferrin. ExcelTaq™ Blood Direct DNA Polymerase is compatible with most anticoagulants, such as citrate, EDTA, and heparin (Fig. 1). The ExcelTaq™ Blood Direct DNA Polymerase includes a pair of Positive Control Primers (CCR5) that are compatible with primate blood samples.

Free-circulating nucleic acids, such as tumor-specific extracellular nucleic acid fragments and mRNAs in the blood or fetal nucleic acids in maternal blood, are present in serum or plasma usually as short fragments, <1000bp(Nucleic Acid). The HiPure Circulating Nucleic acid Micro Kit enables efficient purification of these circulating nucleic acids from human plasma, serum, or urine. Samples can be either fresh or frozen (provided that they have not been frozen and thawed more than once). Free-circulating cell-free DNA, RNA or viral nucleic acids are eluted in Nuclease Free Water, ready for use in amplification reactions or storage at -30 to -15°C. Purified nucleic acids are free of proteins, nucleases, and other impurities.

This product is based onsilica column purification. The sample is lysed and digested with lysate andprotease, DNA is released into the lysate. Transfer to an adsorption plate andfilter column. Nucleic acid is adsorbed on the membrane, while protein is notadsorbed and is removed with filtration. After washing proteins and otherimpurities, Nucleic acid was finally eluted with low-salt buffer.

Storage and Stability
Proteinase K, Carrier RNAshould be stored at 2-8°C upon arrival. However, short-term storage (up to 12weeks) at room temperature (15-25°C) does not affect their performance. Theremaining kit components can be stored dry at room temperature (15-25°C) andare stable for at least 18 months under these conditions. The entire kit can bestored at 2-8°C, but in this case buffers should be redissolved before use.Make sure that all buffers are at room temperature when used.
