Test Principle
The technological basis for the GenLine tests is the polymerase chain reaction (PCR) combined with lateral flow Tests.
Labelled specific primers are used to amplify specific DNA fragments. In addition to the target gene, a control gene, which is also present in the PCR mixes, is amplified in order to make sure that the PCR process works properly.
The resulting PCR products carry the labels of the incorporated primers.
In a second part of the test, the created PCR products are detected by a lateral flow Test Strip. A “molecular sandwich” is formed and becomes visible as a line on the test Strip.
Brief Instructions
The entire procedure of the test can be divided into 3 different steps.
1.The PCR reagents and the samples are prepared.
2.After the addition of the sample to the PCR reagents, the PCR is started.
3.The resulting PCR products are detected by a simple lateral flow test Strip

Human Immunodeficiency Virus (HIV) is a retrovirus which destroys essential cells in the human immune system, such as CD4+ lymphocytes (a type of T cell), macrophages and dendritic cells. Infection with HIV therefore leads to an impaired immune system and increased susceptibility to opportunistic cancers and infections. Due to the mutating nature of HIV and damage to the T cells, the immune system is unable to fight off infections and disease. Without treatment, HIV can progress to Acquired Immunodeficiency Syndrome (AIDS). The rate of HIV progression to AIDS depends on a number of different factors including viral, host and environmental factors. However, with adherence to antiretroviral therapy (ART), many individuals with HIV will never progress to AIDS, having near normal life expectancy. Transmission of HIV is through direct contact with infected bodily fluids, either sexually; by the sharing of blood-contaminated needles or instruments; maternally through childbirth or breastfeeding; or via blood transfusion/organ transplant.

Storage Conditions and Product Stability All kit components can be stored for 1 year after the date of production without showing any reduction in performance.
All kit components should be stored at -20°C upon arrival. Repeated thawing and freezing (> 2 x) of the Master Mix and Positive Control should be avoided, as this may affect the performance of the assay. If the reagents are to be used only intermittently, they should be frozen in aliquots.
HiPure DNA Clean Up 96 Kit uses proprietary chemistry and HiPure technology to recover DNA Fragments between 60bp-10kbp with yields exceeding 80%. DNA is suitable for ligations, PCR, sequencing, restriction digestion, or various labeling reactions. In addition, this kit can be also used to recover DNA directly from enzymatic reactions such as PCR and enzyme digestion reactions.

Principle
The HiPure system uses a simple bind-wash-elute procedure. Gel slices are dissolved in a buffer containing a pH indicator, allowing easy determination of the optimal pH for DNA binding, and the mixture is applied to the column. Nucleic acids adsorb to the silica-gel membrane in the high-salt conditions provided by the buffer. Impurities are washed away and pure DNA is eluted with a small volume of low-salt buffer provided or water, ready to use in subsequent applications.

Storage and Stability
The Kit can be stored dry at room temperature (15-25°C) and are stable for at least 18 months under these conditions. If any precipitates form in the buffers, warm at 37℃ to dissolve.
