This kit features a rapid, high-throughput 96-well format for isolating high yield and high quality DNA from saliva. The purified saliva DNA is compatible with any number of downstream applications including PCR, qPCR, Southern Blot analysis, sequencing, microarray analysis and more. This kit is also compatible with Norgen’s Saliva DNA Collection and Preservation Devices (Cat. RU49000). It is fully compatible to both vacuum and centrifugation protocols.
Background
Human genomic DNA found in saliva can be used in various applications in diagnostics including the detection of biomarkers to diagnose a disease, follow the disease’s progress or monitor the effects of a particular treatment. Isolation of DNA from saliva for diagnostics has become an attractive alternative to isolation from blood or tissue due to the fact that sample collection is non-invasive.
Storage Conditions and Product Stability The kit contains ready-to-use Proteinase K which is dissolved in a specially prepared storage buffer. The Proteinase K should be stored at room temperature or 4°C. All other solutions should be kept tightly sealed and stored at room temperature (15 – 25°C). This kit is stable for 1 year after the date of shipment.

Use in combination with Attogene Algae DNA isolation kit
Attogene’s PCR kit for Microcystin is designed for the In vitro analysis of the MycE gene region responsible for assembling part of the Microcystis peptide. The MycE gene region specific primer and probe mix is provided to be detected through the FAM channel on a qPCR machine. A sample of algae is obtained and washed to extract a clean algal gDNA sample. A reaction mixture is assembled from primers, probe, master mix, and gDNA samples as required. The qPCR machine of choice is set up and loaded as needed and the mixture undergoes PCR amplification. The Primer mix provided exploits the Taq polymerase to amplify the gene region of interest; while the DNA probe mixture is cleaved during amplification to release its FAM fluorophore. The resulting FAM release can be detected on a variety of qPCR platforms.

Plasma/Serum cell-free circulating DNA (cfc-DNA) has the potential to provide biomarkers for certain cancers and disease states as well as fetal DNA in maternal blood. Currently, significant advancements are being made in utilizing cfc-DNA as biomarkers for the early diagnosis, prognosis and monitoring of therapy for several cancer types and autoimmune diseases. Cell-free mitocondrial DNA (cfmtDNA) is also under investigation for its clinical significance. This cfc-DNA is usually present as short fragments of less than 1000 bp. In addition, cell-free fetal DNA has been widely used as a non-invasive method for prenatal diagnosis including early identification of fetal sex, genetic studies for families at high risk for inherited genetic disorders, screening for Rhesus factor, screening for aneuploidy and identification of preeclampsia.

Storage Conditions and Product Stability All buffers should be kept tightly sealed and stored at room temperature (15-25°C) for up to 2 years without showing any reduction in performance. Norgen’s Plasma/Serum Cell-Free Circulating DNA Purification Kits contain ready-to-use Proteinase K solution, which is dissolved in a specially prepared storage buffer. The Proteinase K is stable for up to 2.5 years after delivery when stored at room temperature. To prolong the lifetime of Proteinase K, storage at 2–8°C is recommended.
