Product overview
Protease K is a serine protease with high enzymatic activity and wide substrate specificity. It can preferentially decompose ester and peptide bonds adjacent to the ends of hydrophobic amino acids, sulfur-containing amino acids and aromatic amino acids C, and is often used to degrade proteins to produce short peptides. It has the typical catalytic triad Asp39-His69-Ser224 characteristic of serine proteases and two Ca2+ binding sites around the active center increase its stability, allowing it to maintain high enzyme activity under a wider range of conditions.

Adsorption Mechanism
Based on the negatively charged DNA skeleton, it has a high affinity for positively charged glass fibers. In high salt and ethanol solutions, DNA/RNA binds to glass fiber and interacts with hydrophilic matrix on silica through hydrogen bond. DNA/RNA is tightly bound. All pollutants can be removed by washing solution. At high salt concentration, nucleic acids selectively bind to silicagel membrane, while other pollutants, mainly proteins, are removed by membrane washing.

All kit components should be stored at -20°C upon arrival.
Once reconstituted, repeated thawing and freezing (>2 times) of the Master Mix and Positive Control should be avoided, as this may affect the performance of the assay. If the reagents are to be used only intermittently, they should be frozen in aliquots.
Each kit is provided with 4 tubes of 2X PCR Master Mix and each tube is enough to run 25 reactions. It is not necessary to reconstitute all Master Mix tubes at once. The Master Mix tubes can be reconstituted as and when needed.
All kit components can be stored for 2 years after the date of production without showing any reduction in performance.
