The BK virus is a member of the polyomavirus family. It has been suggested that this virus may be transmitted through respiratory fluids or urine, since infected individuals periodically excrete virus in the urine. BK viral infections are typically asymptomatic in healthy individuals, however very mild symptoms may appear including mild respiratory infections and fever. Infections with BK virus in immunocompromised or immunosupressed patients are much more severe and may involve renal dysfunction. In fact, in kidney transplant patients the immunosupressive drugs required for the transplant may allow the virus to replicate within the graft, resulting in a disease called BK virus nephropathy (BKVN). The JC virus is a type of human polyomavirus and is very common in the general population, infecting 70 to 90% of humans. Most people acquire JCV in childhood or adolescence. Typically the infection is subclinical and no of consequence in individuals with healthy immune systems. The initial site of infection may be the tonsils or the gastrointestinal tract, and the virus then remains latent in the gastrointestinal tract. JCV can also infect the tubular epithelial cells in the kidneys, where it continues to reproduce, shedding virus particles in the urine. Also, JCV can cross the blood-brain barrier into the central nervous system. JCV is known to cause the usually fatal progressive multifocal leukoencephalopathy (PML) by destroying oligodendrocytes in the brain in immunodeficient or immunosuppressed individuals. The JC and BK viruses are very similar, with their genomes sharing 75% homology. It is however important to differentiate between the viruses due to the differences in pathology and especially the invariably fatal outcome of PML which is only caused by the JC virus.

All kit components should be stored at -20°C upon arrival. Repeated thawing and freezing (> 2 x) of the Master Mix and Positive Control should be avoided, as this may affect the performance of the assay. If the reagents are to be used only intermittently, they should be frozen in aliquots.

This kit provides a convenient and rapid method to purify total RNA from small amounts of stool samples. All types of stool samples can be processed with this kit, including animal fecal samples, manure and samples collected using Norgen’s Stool Nucleic Acid Collection and Preservation Tubes (Cat. 45660). The kit removes all traces of humic acids using rapid and simple spin column procedures. Bead tubes are also provided for effective homogenization of stool. The kit purifies all sizes of RNA, from large mRNA and ribosomal RNA down to microRNA and small interfering RNA. Both host and microbial RNA is recovered. The protocol does not rely on the use of phenol or chloroform, thereby providing a user friendly procedure and allowing high-throughput analysis. The purified RNA is of the highest integrity, and can be used in a number of downstream applications including real time PCR and reverse transcription PCR for gene expression analysis. The procedure can be completed in approximately 30 minutes for 10 samples.

1. User-Friendly Design: The chamber lid and tabletop are covered with heat-insulating resin materials, providing both protection against burns and an aesthetically pleasing appearance. The upward-opening flip-top door saves space, while the rounded corners prevent injury. The specially designed steam discharge and collection system minimizes steam burn risks and enhances operational convenience.
2. Interlock Mechanism and Safety Protection: The safety interlock system is designed to be more sensitive and secure. The chamber lid cannot be opened if there is pressure or steam inside the chamber. The lid can only be opened when the pressure gauge reads “0” MPa. The sterilizer will not start heating if the chamber lid is not properly locked. These features enhance the overall safety of the machine.
3. Over-temperature and Heating Protection: The system continuously monitors the temperature inside the sterilization chamber. If the temperature becomes too high or heating is abnormal, power will be automatically cut off to prevent overheating.
4. Automatic Fault Detection System: The machine performs an automatic self-check when powered on. If a fault occurs during operation, an audible alarm will sound, and a fault code will be displayed.
5. Lid Closure Check System: The system automatically detects whether the chamber lid is securely locked. If the lid is not properly locked, the sterilizer will not start.
6. Leakage, Over-current, and Short-Circuit Protection: If there is leakage in the heating elements or other components, or if over-current or short-circuit conditions occur, the system will automatically cut off power to prevent user injury.
7. Water Shortage and Independent Dry-Burning Protection: Equipped with a low water level sensor and independent dry-burning protection device, the system can monitor the water level in the chamber in real-time to prevent dry burning and avoid false readings. If the low water level sensor fails, the overheat and dry-burning protection system will automatically activate, cut off power to the heating ring, and sound an alarm to ensure safety.
8. Dual Pressure Protection: The system includes both a mechanical safety valve and an electronic over-pressure protection system. If pressure becomes abnormal, the system will relieve pressure and cut off power while sounding an alarm.
9. Back end Safety Testing: The system can be periodically tested for safety and protective measures.
10. Over-pressure Testing: Regular testing ensures that the instrument’s over-pressure protection system can automatically cut off power or that the safety valve can open to relieve pressure.

This kit provides a rapid method for the high throughput isolation and purification of total RNA, DNA and proteins sequentially from a single sample of cultured animal cells, small tissue samples, blood, bacteria, or yeast. The kit employs two columns: 1) for gDNA purification and 2) for RNA purification utilizing Norgen’s resin columns (superior for the binding of all RNA sizes including miRNA). The proteins are also purified on the second column after RNA elution. The proteins are eluted in buffer and are ready for downstream application without any further clean up required.
