Norgen’s HPV (High and Low Risk) TaqMan PCR Lyophilized Kit is designed for the detection of HPV High and Low Risk specific DNA in a real-time PCR based on the use of TaqMan® technology. The lyophilized format is designed to ship the kit at ambient temperature.

All kit components should be stored at -20°C upon arrival.
Once reconstituted, repeated thawing and freezing (>2 times) of the Master Mix and Positive Control should be avoided, as this may affect the performance of the assay. If the reagents are to be used only intermittently, they should be frozen in aliquots.
All kit components can be stored for 2 years after the date of production without showing any reduction in performance.

Description
The ExcelDye™ 6× DNA Loading Dye (Tri-Color) is pre-mixed buffer for tracking the DNA sample during the electrophoresis on agarose or polyacrylamide gels. It contains three dyes (Xylene cyanol FF, Bromophenol blue, Orange G) for tracking the DNA migration. The Xylene cyanol FF, Bromophenol blue and Orange G migrate at approximately 800 bp, 150 bp and 30 bp on a standard 2% TAE agarose gel respectively (4,000 bp, 500 bp and 50 bp on 1% TAE agarose gel respectively). The included glycerol keeps the DNA at the bottom of the well and the presence of EDTA chelates divalent metal ions to prevent the process of metal-dependent nuclease.

MagZol Reagent, included in the kits, is a monophasic solution of phenol and guanidine thiocyanate, designed to facilitate lysis of samples and inhibit RNases. The high lysis efficiency of the reagent and the subsequent removal of contaminants by organic phase extraction enable extracting up to 200μg total purified RNA from less than 100mg animal, plant, fungal samples,bacteria, cells and other samples.

Main Functions
Isolation total RNA (include miRNA) from 100mg lipid tissue, tissue, cell, plant, body fluids using columns and MagZol reagent
Applications
RT-PCR, qRT-PCR, Northern hybridization, second generation sequencing
Tissue samples (10-100mg) are homogenized in MagZol Reagent. After addition of chloroform, the homogenate is separated into aqueous and organic phases by centrifugation. The upper, aqueous phase is extracted, and ethanol is added to provide appropriate binding conditions. The sample is then applied to the spin column, where the total RNA (up to 100µg) binds to the membrane and phenol and other contaminants are efficiently washed away. High-quality RNA is then eluted in 30-100µl of RNase-free water.

Storage and Stability
MagZol Reagent should be stored at 2-8°C upon arrival. However, short-term storage (up to 12 weeks) at room temperature (15-25°C) does not affect its performance. The remaining kit components can be stored at room temperature (15-25°C) and are stable for at least 18 months under these conditions.
