Isothermal nucleic acid Principle Summary

This kit is based on a rapid nucleic acid amplification technology at room temperature and constant temperature:at room temperature and constant temperature(generally 39℃~42℃),reverse transcriptaseuses specific primers and template RNA tosynthesize cDNA strands,and binds the auxiliary protein and single strand with the help of the protein, the recombinase and the primer form a complex; perform a homology search and bind the target homology domain, at this time a D-loop region is formed at the homology positionand strand exchange begins; accompanied by the recombinase from the complex upon dissociation,the polymerase also bindsto the 3′ end of the primer,initiating chainextension.It is suitable for laboratory-level RNA amplification and RNA amplification for other detection purposes.

Isothermal nucleic acid Applications

Suitable for RNA isothermal rapid amplification kit(Basic type)

Primer: Require pair of nucleotide primers with the length of 25-35 bp.

RNA basic kit reaction temperature is 39 to 42℃ and time is 5-20 minutes.

1/ Please avoid nucleic acid contamination and set blank control during reaction due to the high sensitivity of the kit.

2/ Please take out the required quantity of MIRA reaction units for the experiment, and put the rest under storage conditions when performing the experiment.

Introduction

Usages:

For pre-enrichment of Salmonella spp.

Principle:

Peptone provide carbon and nitrogen sources to meet the needs of bacterial growth; sodium chloride maintains osmotic equilibrium; potassium dihydrogen phosphate and disodium hydrogen phosphate as buffer.

Formulation(per liter):
Peptone: 10g
Sodium chloride:5g
Disodium hydrogen phosphate:3.5g
Potassium dihydrogen phosphate: 1.5g
Final pH7.2 ± 0.2

How to use:
1. Suspend 20g of product, adding 1L of distilled or deionized water, heated to boiling stirring until completely dissolved, dispensing into flask, autoclave at 121 ℃ for 15min, set aside.
2.Diluted and treated samples.

The ExcelTaq™ 5X PCR Master Dye Mix is a ready-to-use mixture for amplifying targeted DNA fragments. It is designed to serve as ready-to-use master mix for virtually all PCR applications. The mixture contains all components for PCR with the exception of template and primers. This not only saves valuable time in the laboratory, but also reduces the number of pipetting and reagent handling errors. The PCR Master Dye Mix is supplied as a 5X concentrated ready-to-use mix, that is a mixture of recombinant Taq DNA polymerase, reaction buffer, MgCl₂, dNTP, enzyme stabilizer and PCR-friendly loading dye solution containing tracking dye (Bromophenol blue) enabling efficient amplification of template in PCR and allows the user to prepare a PCR reagent – loading dye master mix conveniently.
